Introduction: The effect of diabetes induced in mice on the cytokines interferon (IFN)-γ, interleukin (IL)-1β, and tumor necrosis factor (TNF)-α was studied. As (IFN)-γ, interleukin IL-1β, in addition to tumor necrosis factor (TNF)-α each up regulate the expression of major histocompatibility complex (MHC). (IFN)-γ, interleukin IL-1β and tumor necrosis factor (TNF)- α are therefore considered as inflammatory markers. The objective of the present study was to estimate the serum level of (IFN)-γ, interleukin IL-1β, and tumor necrosis factor-alpha (TNF-α) in diabetic mice. As an approach that aims to prove the possible correlation between the level of cytokines and diabetes induced in mice.
Materials and Methods: In a search for the relation of cytokines with Diabetes Mellitus, it deemed of interest to investigate levels of these cytokines in type 1 diabetes. This was done through the detection of serum IFN-γ, IL1-β as well as TNF-α in mice that were treated with Streptozotocin to induce Diabetes. Within 48 hours of its injection into mice, lipopolysaccharide (LPS) induces hyperglycemia and the production of various cytokines.
Results: Diabetes was induced in mice with streptozotocin injection and the level of the cytokines, IFN-gamma, IL1-β and TNF-alpha was measured and correlated to blood levels of glucose, insulin lipid profile, in addition to weight. The serum levels of the three studied cytokines, IFN-γ, IL 1-β and TNF-α were statistically significantly higher among diabetic mice as compared to the control group. Diabetic male mice (M-STZ mice) group showed significantly higher lipid profile as compared to the control group(M-control). Cholesterol level was significantly higher among female control(F-control) as compared to the M-control group. Cholesterol level was significantly higher among diabetic female mice (F-STZ mice) as compared to the F-Control group. Regarding IFN-γ, IL 1-β and TNF-α levels there were significant linear correlations with the glycemic profile (Glucose, insulin) reflected as positive correlation with blood glucose level and negative inversed proportional correlation with insulin level. M-STZ mice group showed significantly higher lipid profile as compared with M-control mice group.
Conclusion: The serum levels of the three cytokines (IFN-γ, IL 1-β and TNF-α) were significantly higher among diabetic mice as compared with control mice.
Regarding IFN-γ, IL 1-β level and TNF-α levels there were significant linear correlations with the glycaemic profile as the IFN-γ, IL 1-β level and TNF-α level had high strong positive correlation with blood glucose level and negative inversed proportional correlation with insulin level. |
