:
MicroRNAs (miRNAs) are small non-coding RNAs that regulate RNA expression either by mRNA degradation or translation inhibition and play an important role in cancer development and progression. However, little is known about the role of miRNAs in leukemogenesis. The purpose of this research is to evaluate the expression pattern of miR-29a in myeloid leukemia, and to study the correlation of miRNA expression with the Philadelphia chromosome abnormalities in CML pateints. Quantitative real-time PCR was used to detect the miRNA expression in 22 bone marrow samples diagnosed with chronic and acute myeloid leukemia in reference to two endogenous control (RNU48, and RNU44). Of all samples tested, miR-29a expression was up-regulated in 27.27% of the cases, down-regulated in 50% of the cases and no changes in 22.2%. In addition, miR-29a expression in AML was significantly downregulated compared to the control. Moreover, For Philadelphia positive patients, miR-29a expression did not change in patients with in B2/a2 mutation. On the other hand, miR-29a was significantly upregulated in 50% of the patients with B3/a2 mutation (3 out of 6) and was significantly downregulated in 33% of the patients with B3/a2 mutation (2 out of 6) and only one patient with B3/a2 (1 out of 6) showed no changes. These data further support the role of miR-29a deregulation in myeloid leukemogenesis and the possibility of regulating miR-29a expression as therapeutic promise for myeloid leukemia in the future.
